What is dasatinib (Sprycel) and how does it work?
Dasatinib is a targeted oral anticancer medication known as a BCR-ABL tyrosine kinase inhibitor (TKI). In certain leukemias, a genetic abnormality called the Philadelphia chromosome produces an overactive BCR-ABL protein that drives uncontrolled cell growth. Dasatinib works by blocking BCR-ABL and several other kinases, slowing or stopping the growth of cancer cells.

How do I take dasatinib (Sprycel)?
Standard dosing is 100 mg once daily for chronic phase CML, and 140 mg once daily for accelerated or blast phase CML and for Ph+ ALL. Dasatinib can be taken with or without food. Swallow tablets whole and do not crush, cut, or chew them. Take at approximately the same time each day. If you miss a dose, take it as soon as you remember unless it is almost time for your next dose; do not take two doses at the same time. Always follow your oncologist’s specific dosing instructions.
What are the common side effects of dasatinib (Sprycel)?
The most commonly reported side effects of dasatinib include fluid retention (including pleural effusion), diarrhea, headache, fatigue, nausea, rash, and low blood cell counts (myelosuppression). Most people experience some side effects, but severity varies. This is not a complete list. What are serious side effects to watch for with dasatinib?
Serious side effects that require prompt medical attention include severe fluid retention (pleural effusion, pericardial effusion, or pulmonary edema), severe bleeding, QT prolongation, pulmonary arterial hypertension, and severe myelosuppression (very low blood counts). What medications and foods interact with dasatinib?
Dasatinib has several clinically important interactions. Strong CYP3A4 inhibitors (including certain antifungals and some antibiotics) can increase dasatinib levels and raise the risk of toxicity. Strong CYP3A4 inducers (including certain seizure medications and rifampin) can lower dasatinib levels and reduce its effectiveness. Antacids, H2 blockers, and proton pump inhibitors may reduce dasatinib absorption and should be timed or avoided per your prescriber’s guidance. Grapefruit and grapefruit juice can also increase drug levels and should be avoided. Always give your oncologist and pharmacist a full list of medications, supplements, and foods you use.
Can I take dasatinib (Sprycel) if pregnant or breastfeeding?
Dasatinib can harm an unborn baby and is not recommended during pregnancy. Patients of childbearing potential are advised to use effective contraception during treatment and for a period after the last dose. Dasatinib may also pass into breast milk; breastfeeding is not recommended during treatment and for two weeks after the last dose. Discuss your family planning situation with your oncologist before starting or stopping treatment.
